When I tried to build an End Portal in creative, it didn't activate when I placed all the eyes. I tried standing in the middle but it won't teleport me anywhere at all. I tried it on Client and my Server. Minecraft 1.4.4 Vanilla.

Because when you activate the last one, it checks if it's bordering a 3x3 area bordered by 11 other portal border blocks, and it's easier to check one direction than 4. I guess 🙂
But that's at least an explanation that makes sense. I can confirm that End Portals placed in creative mode correctly (as Chalmes states) work.
The orientation of the blocks is critical as per my screenshot. Try replacing the blocks by building them in a circle around you.
Edit: The block that the last eye of ender is placed into must have been placed facing the centre.
