I've just had that on JE 1.16.2 for a second time, but while first time item disappeared forever, this time i paused to search the issue, and when I returned, the item was back in the slot. Odd.

I never got to fix those villagers, unless they get turned into zombie villagers and I cure them again. But new cured zombie villagers don't lose their discounts, so it appears to be a one-time issue due to the update.
