You cannot remove silk touch from an item by trying to put fortune on it. When you try to combine two mutually exclusive enchantments, it will not put anything of the two enchantments on it.

The only way would be to put a pickaxe with fortune in the first slot of the anvil, then the silk touch pickaxe in the second slot of the anvil. It requires you to pay the enchant cost of adding all of the enchantments except the silk touch to the fortune pickaxe.
